General description

TMPK, also known as dTMP kinase, and encoded by the gene DTYMK/CDC8/TMPK/TYMK, is a key enzyme for pyrimidine synthesis that catalyzes the phosphorylation of thymidine 5′-monophosphate (dTMP) to form thymidine 5′-diphosphate (dTDP). TMPK has important functions in cell proliferation and DNA replication, and the synthesis of dTDP is unique because it requires TMPK. All other dNDPs are directly produced by ribonucleotide reductase (RNR). Thus, TMPK is well recognized as a potential drug target for many diseases and disease agents including viruses. TMPK is currently a target of anti-HIV nucleoside therapy, and TMPK is also a validated target for antibiotic development against resistance strains of bacteria and parasites, and research demonstrates that it may also be a good target for the anticancer agent doxorubicin cancer cells. Expressed by virtually all cells it is localized to cell cytoplasm with only traces found in Golgi, mitochondria and nuclei.
